Abstract

Quantifying antibodies against Plasmodium vivax enables the detection of both recent and past P. vivax exposure and may help identify individuals with clinically silent hypnozoite infections. Conversely, the absence of antibodies may support the confirmation of elimination. This protocol describes a multiplexed Luminex-based assay that measures total IgG antibody responses against eight validated P. vivax serological exposure markers (SEMs) using plasma or dried blood spot samples. The workflow incorporates cost-conscious adaptations, minimal sample volume, and flexible sample types to support scalable surveillance and serology-based test-and-treat strategies in resource-limited settings.
